Florida pompano (Trachinotus carolinus) are a species of growing interest for commercial aquaculture. Effective health monitoring is crucial to the successful growout of the species, and prophylactic and therapeutic use of chemicals and antibiotics has been the traditional strategy for promoting stock health. However, concerns about antimicrobial resistance, chemical residues in seafood products and the environment, and resultant immunosuppression have prompted the industry to identify alternative management strategies, including supplementation with prebiotics, probiotics, and combinations of both (synbiotics). The objectives of this study are to determine and compare hematological, plasma biochemical, and plasma protein electrophoresis data of synbiotic-supplemented (β-glucan and Pediococcus acidilactici) and non-supplemented Florida pompano. Reference intervals for blood analytes are provided for both groups and for subgroups (females, males, large, and small fish) where statistically significant results exist. There are no differences between the hematological and plasma biochemistry analytes between the supplemented and control groups, except for blood urea nitrogen and carbon dioxide, indicating a possible effect of synbiotic supplementation on gill function and osmoregulation. Sex-related and size-related differences are observed within each of the control and supplemented groups; however, biometric measurements do not strongly correlate with blood analytes. These data represent baseline hematological and plasma biochemical data in the Florida pompano and indicate the safety of synbiotic supplementation in this commercially important species. This study serves to further the commercialization of Florida pompano by providing blood analyte reference intervals for health monitoring in the aquaculture setting.  相似文献

In molecular phylogenetics, mode of analysis is typically the primary reason cited for obtaining different topologies. However, sequence alignment is as important in determining topology as analytical method or optimality criterion, particularly for length-variable sequences. We investigate the effect of alignment strategy and parameters in an analysis of length-variable 18S rDNA sequences from 41 oceanic squids (Decapodiformes), focusing particularly on the impact of including variable regions on the resulting topology. Decapodiformes are an enigmatic group of molluscs whose evolutionary relationships remain unclear because standard molecular analyses have continued to produce poorly supported or poorly resolved trees, possibly due to elevated levels of molecular evolution. We find that alignment strategy (Clustal X versus POY) has a greater affect on topology than does parameter choice. Some groups were robust to shifts in methodology, including cuttlefishes (Sepiidae), bobtail squids (Sepiolidae) and pygmy squids (Idiosepiidae). No alignment strategy supported the monophyly of Oegopsida, although Myopsida did consistently form a monophyletic group. The variable regions consistently provided more well-supported and well-resolved trees than the conserved regions alone, indicating the phylogenetic importance of including length-variable data. © The Willi Hennig Society 2007.  相似文献

The routine production and storage of frozen doughs are still problematic. Although commercial baker's yeast is highly resistant to environmental stress conditions, it rapidly loses stress resistance during dough preparation due to the initiation of fermentation. As a result, the yeast loses gassing power significantly during storage of frozen doughs. We obtained freeze-tolerant mutants of polyploid industrial strains following screening for survival in doughs prepared with UV-mutagenized yeast and subjected to 200 freeze-thaw cycles. Two strains in the S47 background with a normal growth rate and the best freeze tolerance under laboratory conditions were selected for production in a 20-liter pilot fermentor. Before frozen storage, the AT25 mutant produced on the 20-liter pilot scale had a 10% higher gassing power capacity than the S47 strain, while the opposite was observed for cells produced under laboratory conditions. AT25 also retained more freeze tolerance during the initiation of fermentation in liquid cultures and more gassing power during storage of frozen doughs. Other industrially important properties (yield, growth rate, nitrogen assimilation, and phosphorus content) were very similar. AT25 had only half of the DNA content of S47, and its cell size was much smaller. Several diploid segregants of S47 had freeze tolerances similar to that of AT25 but inferior performance for other properties, while an AT25-derived tetraploid, TAT25, showed only slightly improved freeze tolerance compared to S47. When AT25 was cultured in a 20,000-liter fermentor under industrial conditions, it retained its superior performance and thus appears to be promising for use in frozen dough production. Our results also show that a diploid strain can perform at least as well as a tetraploid strain for commercial baker's yeast production and usage.  相似文献

Undecaprenyl pyrophosphate synthase (UPPS) catalyzes the consecutive condensation reactions of eight isopentenyl pyrophosphate (IPP) with farnesyl pyrophosphate (FPP) to generate C(55) undecaprenyl pyrophosphate (UPP). In the present study, site-directed mutagenesis, fluorescence quenching, and stopped-flow methods were utilized to examine the substrate binding and the protein conformational change. (S)-Farnesyl thiopyrophosphate (FsPP), a FPP analogue, was synthesized to probe the enzyme inhibition and events associated with the protein fluorescence change. This compound with a much less labile thiopyrophosphate shows K(i) value of 0.2 microm in the inhibition of Escherichia coli UPPS and serves as a poor substrate, with the k(cat) value (3.1 x 10(-7) s(-1)) 10(7) times smaller than using FPP as the substrate. Reduction of protein intrinsic fluorescence was observed upon addition of FPP (or FsPP) to the UPPS solution. Moreover, fluorescence studies carried out using W91F and other mutant UPPS with Trp replaced by Phe indicate that FPP binding mainly quenches the fluorescence of Trp-91, a residue in the alpha3 helix that moves toward the active site during substrate binding. Using stopped-flow apparatus, a three-phase protein fluorescence change with time was observed by mixing the E.FPP complex with IPP in the presence of Mg(2+). However, during the binding of E.FsPP with IPP, only the fastest phase was observed. These results suggest that the first phase is due to the IPP binding to E.FPP complex, and the other two slow phases are originated from the protein conformational change. The two slow phases coincide with the time course of FPP chain elongation from C(15) to C(55) and product release.  相似文献

Laboratory experiments were performed with adult female Macrolophus caliginosus Wagner (Heteroptera: Miridae) at 22ºC on bean plants to determine the functional response towards whiteflies, as well as the preference and switching capacity between the two prey species: whiteflies and spider mites. Predation of females presented with first instars of Trialeurodes vaporariorum Westwood (Homoptera: Aleyrodidae) was of a Type III functional response. The observed maximum predation was approximately 75 first instars at high prey densities within a 24-h period. The preference of M. caliginosus females between eggs of T. vaporariorum and Tetranychus urticaeKoch (Acarina: Tetranychidae) changed with the ratio of offered prey. The preference for T. vaporariorum eggs increased non-linearly with increasing proportions of this prey type. The average maximum predation of whitefly and spider mite eggs were approximately 166 and 111 eggs per day, respectively, at the highest ratio of the two preys. The proportion of M. caliginosus females found on the test plants at the end of the experiment increased with prey density suggesting that this mirid spends more time in areas with high prey density. Macrolophus caliginosus females are voracious predators of eggs and first instars of T. vaporariorum as well as of spider mite eggs and may thus be a valuable addition to existing methods of biological control of T. vaporariorum and T. urticae.  相似文献

A collaborative international project funded by the European Union's INCO-DC programme is undertaking limnological, fish biological, environmental and socio-economic research in five tropical lakes and reservoirs in Sri Lanka, Thailand and the Philippines over the period 1998–2001. The aim is to determine their trophic structure and their capacity to sustain both their existing fisheries and present and future aquaculture. In some cases, these activities could potentially be expanded for the benefit of rural communities and of the local market within the bounds of social and environmental sustainability. This paper describes the concepts and methods involved in this innovative multidisciplinary project which aims to integrate limnological, fisheries and socio-economic issues in a comparative approach involving Asian and European research teams.Deceased.  相似文献

Insulin and insulin-like growth factor 1 (IGF-1) receptor signaling pathways differentially modulate cardiac growth under resting conditions and following exercise training. These effects are mediated by insulin receptor substrate 1 (IRS1) and IRS2, which also differentially regulate resting cardiac mass. To determine the role of IRS isoforms in mediating the hypertrophic and metabolic adaptations of the heart to exercise training, we subjected mice with cardiomyocyte-specific deletion of either IRS1 (CIRS1 knockout [CIRS1KO] mice) or IRS2 (CIRS2KO mice) to swim training. CIRS1KO hearts were reduced in size under basal conditions, whereas CIRS2KO hearts exhibited hypertrophy. Following exercise swim training in CIRS1KO and CIRS2KO hearts, the hypertrophic response was equivalently attenuated, phosphoinositol 3-kinase (PI3K) activation was blunted, and prohypertrophic signaling intermediates, such as Akt and glycogen synthase kinase 3β (GSK3β), were dephosphorylated potentially on the basis of reduced Janus kinase-mediated inhibition of protein phosphatase 2a (PP2A). Exercise training increased peroxisome proliferator-activated receptor gamma coactivator 1-alpha (PGC-1α) protein content, mitochondrial capacity, fatty acid oxidation, and glycogen synthesis in wild-type (WT) controls but not in IRS1- and IRS2-deficient hearts. PGC-1α protein content remained unchanged in CIRS1KO but decreased in CIRS2KO hearts. These results indicate that although IRS isoforms play divergent roles in the developmental regulation of cardiac size, these isoforms exhibit nonredundant roles in mediating the hypertrophic and metabolic response of the heart to exercise.  相似文献

Body water is a major element of the cold-hardiness strategies observed in ectothermic animals, in particular in freezing avoidant species for which body ice formation is lethal. Here, we investigate the relationships, in terrestrial snails, between the temperature of crystallisation (Tc) and body water (water mass and water content), shell shape, geographic and climatic distribution, taking into account phylogenetic inertia. Phylogenetic relationships among 31 species from 13 different families of terrestrial Gastropods were studied using 28S rRNA nuclear and COI mitochondrial sequence data, together with species-specific traits. Our results provide evidence for clear relationships between Tc and absolute/relative body water: smaller species with lower water content tended to be characterized by colder temperatures of crystallisation, although some exceptions were noticeable. Environmental conditions do not appear to affect Tc significantly, as well as shell shape which is however correlated with water content. This study confirmed that supercooling ability in land snails is size-constrained, with consequences on cold-hardiness strategies.  相似文献
